Capsule procedure

Capsule staining is used to detect capsules or slime layers. During this experiment a nigrosin stain was used which looks like a black ink. First we smeared our bacteria onto the edge of a slide along with a nigrosin stain. We proceeded by using another slide to smear the stain and bacteria across the entire surface of the original slide. After letting the stain dry, we covered it with crystal violet and washed off the excess before observing our results under a microscope. The smear indicated that we did not have a capsule surrounding our bacteria.
