Maintenance and storage of stock culture.

         Today we first learned how to preserve our bacteria for a long period of time by freezing. Freezing the bacteria alone may cause ice-crystals to form. Therefore, glycerol is added to the medium which preserves bacteria cells  in freezing conditions. The bacteria then can be stored in -20 degrees-70 degrees C temperatures for up to five years. 



Our procedure began by filing a small microcentrifuge with 350 micrometers broth, and  150 micrometers glycerol to have an overall ratio of 70% broth, and 30% glycerol. Before we placed the medium into the freezer, we shacked the contents vigorously.
